The CRISPR-Cas9 system is a revolutionary technology that has transformed the field of genome editing. The Cas9 nuclease is a key component of this system, and its ability to specifically bind to and cleave target DNA sequences has made it a powerful tool for researchers. In recent years, novel Cas9 nucleases have been developed that have improved upon the original SpCas9, making it even more precise and safer.
In this presentation, we will discuss the development and characteristics of one novel Cas9 nuclease. Because of its high intrinsic specificity, we named the new enzyme Specific Off-Target/ON-target (SpOT-ON) discriminator. We will present the in vitro and in vivo data to support its high specificity and gene editing outcomes that lead to a reduction in chromosomal translocations. Our results suggest that SpOT-ON is a valuable alternative to SpCas9 for research and clinical use, and has the potential to significantly impact the field of genome editing.
