Synthetic nanobodies (sybodies) have long been known to be versatile tools to support structure determination of flexible targets by stabilising a desired conformation or aiding crystal contact formation as well as increasing the size of the target for cryo-EM. Prior to this, sybodies also greatly benefit the expression and purification of flexible and unstable protein targets, allowing the generation of high quality protein reagents for biochemical and biophysical assays as well as small molecule screening efforts. In this talk we highlight the impact of sybodies in protein science and share some examples from recent protein production campaigns.
