High-content imaging and screening with automated analysis made easy using the Hermes® imaging system from IDEA Bio-Medical

Objective

The Hermes WiScan® automated microscope coupled with versatile and fully automated WiSoft® Athena software enables the development of quantitative, sub-cellular imaging assays for high-throughput phenotypic profiling.

Here, we present case studies where the Hermes system enabled the acquisition of large datasets to probe cellular responses to chemicals, viral infection and protein-protein interactions with robust and reliable statistics.

In each instance, researchers acquired tens of thousands of multicolour fluorescence images using the Hermes WiScan® hardware.  Working closely with Idea Bio-Medical, they designed custom-made image analysis packages to automate their quantification needs.

Researchers identified several novel interactions through their images and analysis:
- Nuclear translocation inhibition of intracellular immune signals by HIV-1 virus as a requirement for infection;
- Alteration in actin-dependent protein-protein interactions by bacterial infection leading to the innate immune response;
- Quantified coupling of drug cytotoxicity with alterations in protein expressions for chemotherapy and epigenetics.
